[目的]应用免疫组化法,观察茶多酚口服、静脉与局部注射三种给药途径对移植性小鼠血管内皮生长因子(VEGF)、金属蛋白酶组织抑制因子Ⅱ(TIMP-2)表达影响,进行茶多酚抗肿瘤新生血管生成研究。[方法]建立移植性S180小鼠肿瘤动物模型,随机分茶多酚口服灌胃、局部注射、腹腔注射、人参皂苷Rg3口服灌胃、CTX腹腔注射对照、生理盐水肿瘤模型对照、正常空白对照7组,5d后取肿瘤组织,石蜡包埋、切片,免疫组化染色观察VEGF、TIMP-2表达。[结果]茶多酚口服及局部注射组可明显降低S180荷瘤VEGF表达水平、增加TIMP-2表达水平。[结论]茶多酚口服、局部注射具有抗肿瘤新生血管生成作用。
[Objective] To investigate the effect of oral administration of tea polyphenols (PO), intravenous injection and local injection on the expression of vascular endothelial growth factor (VEGF) and tissue inhibitor of metalloproteinase (TIMP-2) in transplant mice by immunohistochemistry , Polyphenols antitumor neovascularization research. [Method] Tumor animal model of S180 mice was established. The rats were randomly divided into three groups: oral administration of tea polyphenols, local injection, intraperitoneal injection, ginsenoside Rg3 orally, CTX intraperitoneal injection, normal saline control group, normal blank control 7 After 5 days, the tumor tissues were taken and embedded in paraffin. The expression of VEGF and TIMP-2 were observed by immunohistochemical staining. [Result] Oral and topical injection of tea polyphenols could significantly reduce the expression of VEGF in S180 tumor and increase the expression of TIMP-2. [Conclusion] Oral and topical injection of tea polyphenols can inhibit tumor angiogenesis.
