目的:观察地锦草提取物对皮肤癣菌超微结构的影响。方法:参照NCCLS推荐的M38-A方案,将受试药与皮肤癣菌26℃共同培养7d后分离菌体,按常规方法进行样品处理,在扫描电镜和透射电镜下观察其细胞表面形态结构变化与细胞内超微结构变化。结果:扫描电镜下观察地锦草提取物作用于皮肤癣菌后,真菌细胞表面皱缩不平,有严重皱褶、破裂现象;透射电镜下可见,真菌细胞壁不完整,局部有缺损,厚薄不均;细胞膜轮廓不清,局部有破损;胞内细胞器损伤严重,多见空泡化,细胞内成分聚集成电子密度较高的团块。结论:地锦草提取物作用皮肤癣菌后,可使其形态和超微结构发生明显的改变。
OBJECTIVE: To observe the effect of Humulus japonicus extract on ultrastructure of dermatophytes. Methods: According to NCLLS recommended M38-A protocol, the tested drugs were co-cultured with dermatophyte at 26°C for 7 days and the cells were isolated. The samples were processed according to the conventional methods. The cell surface morphology and structural changes were observed under scanning electron microscope and transmission electron microscope. With intracellular ultrastructural changes. Results: Under the scanning electron microscope, the surface of the fungus cells was crumpled and creased when the extracts of Euphorbia japonicum were treated with dermatophytes. Under the transmission electron microscope, the cell wall of the fungi was incomplete, and there were partial defects, and the thickness was not uniform. The cell membrane is unclear in shape and damaged locally; intracellular organelles are severely damaged, vacuoles are more common, and intracellular components aggregate into clumps of higher electron density. Conclusion: Euphorbia decumbens extract can significantly change the morphology and ultrastructure of dermatophytes.
