目的通过体外代谢方法,研究五氯苯酚(pentachlorophenol,PCP)的代谢情况。方法采用肝微粒体和肝S9成分2种体外代谢试剂模拟体内代谢模式,通过优化代谢反应条件,建立PCP、四氯苯醌(Cl4BQ)2种目标物的气相色谱-质谱联用(GC-MS)检测方法,对PCP进行体外代谢研究分析。结果体外代谢分析的最佳反应条件为代谢试剂浓度0.75 mg/m L、代谢反应时间4 h。结论在最优反应条件下分析比较2种试剂对PCP的代谢情况,验证了PCP的代谢产物为Cl4BQ,且在肝微粒体和肝S9成分中的代谢没有显著差异。
Objective To study the metabolism of pentachlorophenol (PCP) by in vitro metabolic method. Methods Metabolic models of in vivo metabolism of liver microsomes and liver S9 were used to optimize the metabolic reaction conditions. Gas chromatography-mass spectrometry (GC-MS) was established for the determination of two target compounds of PCP and chloranil (Cl4BQ) ) Detection methods, in vitro metabolism of PCP research and analysis. Results The optimal reaction conditions for in vitro metabolic analysis were as follows: the concentration of the metabolite was 0.75 mg / m L and the metabolic reaction time was 4 h. Conclusion Under the optimal reaction conditions, the metabolism of PCP was analyzed and compared. The metabolites of PCP were confirmed to be Cl4BQ. There was no significant difference in the metabolism between liver microsomes and liver S9 components.
