目的探讨冠状静脉窦阻塞后,对心肌组织肌钙蛋白Ⅰ(cTnI)和纤维连接蛋白(Fn)的影响。方法健康家犬18只,随机分为对照组、阻塞1 h组和阻塞2 h组,每组6只,对照组不结扎冠状静脉窦,后2组从心外膜在最近冠状静脉窦口处,结扎冠状窦,分别于阻塞后1 h和2 h处死犬。3组均在左心室前壁同一部位取材,行免疫组织化学染色,并进行分析。结果 3组犬实验前后血浆cTnI和Fn含量无明显变化(P>0.05)。与对照组比较,阻塞1 h组和阻塞2 h组心肌组织cTnI表达明显下降,Fn表达明显升高(P<0.01),且阻塞2 h组较阻塞1 h组变化更明显(P<0.01)。结论健康犬冠状静脉窦阻塞后,可导致心肌组织cTnI、Fn呈缺血性改变。
Objective To investigate the effects of coronary sinus occlusion on cardiac troponin Ⅰ (cTnI) and fibronectin (Fn). Methods Eighteen healthy domestic dogs were randomly divided into control group, obstruction 1 h group and obstruction 2 h group, with 6 rats in each group. The control group did not ligate the coronary sinus, , Ligation of the coronary sinus, were killed at 1 h and 2 h after occlusion. All three groups were taken from the same site of anterior wall of left ventricle and immunohistochemically stained. Results There was no significant change in plasma cTnI and Fn levels before and after experiment (P> 0.05). Compared with the control group, the expression of cTnI and the expression of Fn were significantly decreased (P <0.01), and the changes of cTnI in obstructive 1 h group and obstructive 2 h group were more obvious than those in obstructive 1 h group (P <0.01) . Conclusion The occlusion of coronary sinus in healthy dogs can lead to ischemic changes of cTnI and Fn in myocardium.
