利用混合淋巴细胞培养反应(MixedLymphocyteReaction,MLR)研究了传统中药当归(AngelicaSinensisDiels,ASD)来源的两种免疫调节剂当归多糖(ASDP)、当归内酯(ASDE3)的免疫作用机理。结果表明:两者均能明显促进由同种异型抗原刺激的正常小鼠及S180荷瘤小鼠脾细胞增殖(P<0.01),其促进作用与剂量之间呈依赖关系。在一定浓度范围内(16~500μg/ml)对未经活化的脾细胞无诱导增殖作用。选择促进效应较大的药物剂量,培养96h,对活化的T淋巴细胞亚群的变化标记检测,结果显示:ASDP可明显增加L3T+4细胞比例,ASDE3可明显增加L3T+4和Lyt+2细胞的数量。在浓度为250μg/ml时,ASDE3增强细胞毒T细胞的功能,其杀伤活性增加80%。
The immune mechanism of Angelica sinensisDiels (ASD), two kinds of immunomodulators Angelica sinensisDiels (ASD) and Angelica sinensisDiels (ASDE), was studied by mixed lymphocyte reaction (MLR). The results showed that both of them could significantly promote the proliferation of spleen cells of normal mice and S180 tumor-bearing mice stimulated by alloantigen (P <0.01), and their promotion was dose-dependently. In a certain concentration range (16 ~ 500μg / ml) on the non-activated spleen cells did not induce proliferation. The results showed that: ASDP can significantly increase the proportion of L3T + 4 cells, and ASDE3 can significantly increase the number of L3T + 4 and Lyt + 2 cells. At a concentration of 250 μg / ml, ASDE3 enhances cytotoxic T cell function with an 80% increase in cytotoxic activity.
