目的了解青海省2010年3株乙型流感毒株血凝素HA抗原性和基因特性。方法用MDCK细胞分离培养流感病毒,提取流感病毒核酸,采用RT-PCR方法扩增病毒HA基因,纯化产物进行氨基酸序列测定,用MEGA 4.0分析软件对基因种系进行特性分析。结果青海省2010年人群中监测到的为B型Yamagata系毒株,与WHO推荐的疫苗株B/California/01/2009相比,其核苷酸和氨基酸序列未发生明显改变。结论青海省乙型流感病毒优势流行株其抗原性未发生明显改变,提示2010年WHO推荐的乙型流感疫苗对青海省人群有明显的保护效果。
Objective To understand the antigenicity and genetic characteristics of hemagglutinin HA in three strains of influenza B in Qinghai Province in 2010. Methods The influenza virus was isolated and cultured with MDCK cells, and the influenza virus nucleic acid was extracted. The HA gene was amplified by RT-PCR and the amino acid sequence was determined. The characteristics of the gene were analyzed by MEGA 4.0 software. Results The genotype B Yamagata was detected in the 2010 population in Qinghai Province. Compared with the vaccine strain B / California / 01/2009 recommended by the WHO, the nucleotide and amino acid sequence of the strain did not change significantly. Conclusion There is no significant change in the antigenicity of the prevalent strains of influenza B virus in Qinghai Province, which indicates that the influenza B vaccine recommended by the WHO in 2010 has a significant protective effect on the population of Qinghai.
