目的研究miR-451a对人滋养细胞系(HTR-8/SVneo)细胞迁移、侵袭能力的影响。方法以人工合成的寡核苷酸miR-451a模拟物(miR-451a mimics)、miR-451a抑制物(miR-451a inhibitor)分别转染人滋养细胞,过表达与特异性抑制miR-451a;应用逆转录-实时荧光定量聚合酶链反应技术(q RT-PCR)检测转染后miR-451a的表达;应用划痕实验和Transwell小室侵袭实验观察转染前后细胞的迁移和侵袭能力的变化。结果对比空白组及阴性对照组,人滋养细胞转染miR-451a mimics后,miR-451a表达明显升高,细胞迁移、侵袭能力下降;而转染miR-451a inhibitor后,miR-451a表达明显下降,细胞迁移、侵袭能力提高。结论 miR-451a可以抑制人滋养细胞的迁移、侵袭能力。
Objective To investigate the effect of miR-451a on the migration and invasion of human trophoblast cell line HTR-8 / SVneo. Methods Human trophoblasts were transfected with synthetic oligonucleotides miR-451a mimics and miR-451a inhibitor, respectively. Overexpression and specific inhibition of miR-451a were performed. The expression of miR-451a after transfection was detected by reverse transcription-real-time quantitative polymerase chain reaction (q RT-PCR). The changes of cell migration and invasion before and after transfection were evaluated by scratch test and Transwell chamber invasion assay. Results Compared with the blank group and the negative control group, the miR-451a mimics were transfected by human trophoblast cells, the expression of miR-451a was significantly increased and the ability of cell migration and invasion was decreased. However, miR-451a inhibitor significantly decreased the expression of miR- , Cell migration, increased invasive ability. Conclusion miR-451a can inhibit the migration and invasion of human trophoblast cells.
