目的比较实时荧光定量PCR(RT-PCR)对肝细胞癌(HCC)、乙型病毒性肝炎(简称乙肝)患者及健康体检者血浆游离DNA含量及完整性的检测结果,探讨实时荧光定量PCR(RT-PCR)对HCC的诊断价值。方法收集52例HCC患者、58例乙肝患者和60例健康体检者的静脉血标本,采用RT-PCR技术对各组血浆游离DNA中GAPDH、β-actin400、β-actin167含量及其完整性(β-actin400/β-actin167)进行定量检测,结果采用SPSS软件进行统计学分析。结果健康对照组的血浆游离DNAβ-actin167含量和完整性相比明显低于HCC组(Z=–4.328,P=0.000;Z=–3.885,P=0.001)和乙型肝炎病毒(HBV)组(Z=–2.473,P=0.005;Z=–3.295,P=0.013),且HBV组血浆游离DNA完整性明显低于HCC组(Z=–4.836,P=0.002)。结论 RT-PCR检测血浆游离DNA完整性(β-actin400/β-actin167)可作为HCC辅助诊断的分子生物学指标。
Objective To compare the detection results of plasma free DNA content and integrity in patients with hepatocellular carcinoma (HCC), hepatitis B (Hepatitis B) and healthy subjects by real-time fluorescence quantitative PCR (RT-PCR) RT-PCR) diagnostic value of HCC. Methods Fifty-two patients with HCC, 58 patients with hepatitis B and 60 healthy subjects were enrolled in this study. The contents of GAPDH, β-actin400 and β-actin167 in plasma free DNA and their integrity (β -actin400 / β-actin167) for quantitative detection, the results using SPSS software for statistical analysis. Results Compared with the HCC group, the content and the integrity of plasma free DNAβ-actin167 in the healthy control group were significantly lower than those in the HCC group (Z = -4.328, P = 0.000; Z = -3.885, P = 0.001) and the Hepatitis B virus group Z = -2.473, P = 0.005; Z = -3.295, P = 0.013). The plasma DNA integrity of HBV group was significantly lower than that of HCC group (Z = -4.836, P = 0.002). Conclusion The detection of plasma free DNA integrity by RT-PCR (β-actin400 / β-actin167) can be used as a molecular biomarker for the diagnosis of HCC.
