以大麦品种‘花30’小孢子培养来源单倍体植株作为起始材料,分别对其丛生芽诱导、扩繁、生根以及再生植株水培等技术进行了研究。结果表明,添加0.5 mg·L-1噻苯隆(TDZ)的1/2MS培养基适宜丛生芽诱导;添加1.0 mg·L-1 TDZ的1/2MS培养基适宜丛生芽增殖;分别添加400 mg·L-1水解酪蛋白(CH)、20 mg·L-1亚精胺(Spd)和1.0 mg·L-1多效唑(MET)可以促进丛生芽的健壮;添加0.05 mg·L-1α-萘乙酸(NAA)和1.0 mg·L-1 MET的1/2MS培养基适宜生根。水培7~10 d后,90%以上的再生植株正常生长。
The haploid plants derived from microspore culture of ’30’ of barley were used as starting material to study the induction, propagation and rooting of their buds and the hydroponics of regenerated plants. The results showed that 1 / 2MS medium supplemented with 0.5 mg · L -1 TDZ was suitable for shoot induction, and 1/2 MS medium supplemented with 1.0 mg · L -1 TDZ was suitable for proliferation of shoots. 400 mg L-1 hydrolyzed casein (CH), 20 mg · L-1 spermidine (Spd) and 1.0 mg · L-1 paclobutrazol (MET) 1 / 2MS medium with acetic acid (NAA) and 1.0 mg · L-1 MET was suitable for rooting. After hydroponics for 7-10 days, more than 90% of the regenerated plants grow normally.
