目的 研究支架术后不同时间局部食管组织中相关细胞因子的表达状况 ,以及与再狭窄形成的关系。方法 采用免疫组化 (ABC法 )检测支架术后 1、2、4、8周组织中肿瘤坏死因子 α(TNF α)、转化生长因子 β1(TGFβ1)、血小板源性生长因子 (PDGF)、内皮素 1(ET1)的表达及其变化规律。结果 支架术后 1周组织即开始表达TNF α、ET1,术后 2周TNF α、TGFβ1、PDGF、ET1表达均明显增强 ,术后 4周仍有少量TGFβ1表达 ,术后 8周所有细胞因子均极少表达。着色阳性的细胞主要是巨噬细胞、T 淋巴细胞及肥大细胞 ,ET1主要由血管内皮细胞表达。结论 TNF α、TGFβ1、PDGF、ET1作为炎性细胞的表达产物 ,对纤维化及再狭窄的形成具有重要调控作用 ,与支架术后再狭窄的形成存在密切关系。
Objective To study the expression of related cytokines in local esophageal tissue at different time after stenting and the relationship with restenosis. Methods The expression of tumor necrosis factor α (TNFα), transforming growth factor β1 (TGFβ1), platelet derived growth factor (PDGF) and endothelium of tumor tissue were detected by immunohistochemistry (ABC method) Expression of ET1 and its variation. Results The expression of TNFα and ET1 began to expire at 1 week after operation. The expressions of TNFα, TGFβ1, PDGF and ET1 were significantly increased 2 weeks after operation. A small amount of TGFβ1 remained at 4 weeks after operation. All the cytokines Very rarely expressed. Pigmented positive cells are mainly macrophages, T lymphocytes and mast cells, ET1 is mainly expressed by vascular endothelial cells. Conclusion TNFα, TGFβ1, PDGF and ET1, as the expression products of inflammatory cells, play an important regulatory role in the formation of fibrosis and restenosis, which is closely related to the formation of restenosis after stenting.
