目的研究复方葛根片对衰老模型大鼠肝脏细胞的保护作用。方法 40只大鼠随机分为正常组、模型组(生理氯化钠10ml·kg-1)和复方葛根片低、中、高剂量组[200、400、800mg(生药)·kg-1],每组8只。除正常组外,其他各组大鼠每天9∶00腹腔注射D-半乳糖溶液250mg·(kg·d)-1复制衰老模型,每天15∶00灌胃注射相应药物,连续给药8周。计算肝脏指数,采用TUNEL(TdT-mediated dUTP Nick-End Labeling)法检测衰老大鼠肝脏组织的细胞凋亡情况。结果与正常组比较,模型组大鼠肝脏指数显著增加(P<0.01);肝脏组织中红色荧光明显增多,凋亡指数显著增加(P<0.01);与模型组比较,复方葛根片高、中、低剂量组中大鼠肝脏指数显著减小(P<0.05或P<0.01);肝脏组织中红色荧光明显减少,亮度降低,凋亡指数显著减小(P<0.05或P<0.01)。结论复方葛根片能抑制衰老模型大鼠肝细胞凋亡,减少凋亡细胞的产生,起到保护肝脏细胞的作用。 Objective To study the protective effect of Fuge Pueraria on liver cells of aging model rats. Methods Forty rats were randomly divided into normal group, model group (physiological sodium chloride 10ml · kg-1) and compound Pueraria lobata low, middle and high dose group [200,400,800mg (crude drug) · kg-1] Each group of 8. Except for the normal group, the rats in other groups were intraperitoneally injected with D-galactose solution 250mg · kg -1 at 9:00 every day for replicative senescence. The rats were intragastrically injected with the corresponding drugs at 15:00 for 8 weeks. The liver index was calculated. TUNEL method was used to detect the apoptosis of liver in aged rats. Results Compared with the normal group, the liver index of the model group was significantly increased (P <0.01); the red fluorescence of the liver tissue was significantly increased and the apoptosis index was significantly increased (P <0.01); Compared with the model group, (P <0.05 or P <0.01). In the low dose group, the liver index was significantly reduced (P <0.05 or P <0.01). The red fluorescence of the liver tissue was significantly reduced, the brightness was decreased and the apoptosis index was significantly reduced (P <0.05 or P <0.01). Conclusion Fuge Puerariae can inhibit hepatocyte apoptosis, decrease the production of apoptotic cells and protect liver cells in aging model rats.
