目的:研究大孔树脂富集纯化黄蜀葵花总黄酮的工艺条件。方法:考察8种大孔树脂对黄蜀葵花总黄酮的吸附解吸性能,利用静态和动态吸附解吸试验,筛选出的大孔树脂富集纯化黄蜀葵花总黄酮的最佳工艺。结果:AB-8大孔树脂对黄蜀葵花总黄酮具有较好的吸附解吸能力。AB-8大孔树脂富集纯化黄蜀葵花总黄酮最佳工艺为:吸附液质量浓度为0.75 g生药材/mL,吸附液pH值为4.9,上样流速为1.5 BV/h,上样量为4 BV,洗脱剂为70%乙醇,洗脱剂用量为2.5 BV,洗脱流速为2.0 BV/h。结论:该工艺有效的提高了黄蜀葵花提取物中的总黄酮含量,可用于黄蜀葵花总黄酮的富集纯化。
Objective: To study the conditions of macroporous resin enrichment purification of total flavonoids from Abelmoschus. Methods: The adsorption and desorption properties of total flavonoids from Abelmoschillae macrophylla were studied by using eight kinds of macroporous resins. The optimum conditions for purification of total flavonoids from Abelmoschus chinensis were enriched by static and dynamic adsorption and desorption experiments. Results: AB-8 macroporous resin has good adsorption and desorption ability to total flavonoids of Abelmoschus. The optimum conditions of AB-8 macroporous resin enrichment of purified total flavonoids were as follows: the concentration of absorbent was 0.75 g crude drug / mL, the pH of the adsorption solution was 4.9, the sample flow rate was 1.5 BV / h, 4 BV, 70% ethanol as eluant, 2.5 BV as eluent and 2.0 BV / h as eluent. Conclusion: The process can effectively increase the total flavonoids content of Abelmoschus chinensis flower extract, and can be used for enrichment and purification of total flavonoids of Abelmoschus.
