目的 观察血管内皮生长因子 (vascularendothelialgrowthfactor,VEGF)反义硫代寡脱氧核苷酸 (ASPODN)对裸鼠人胆管癌血管生成和生长的抑制作用。方法 传代培养的胆管癌细胞系QBC93 9,以 5× 10 6 个细胞/ml接种于裸鼠颈背部皮下 ,自 4 8h始局部注射ASPODN和等量的PBS,触及肿瘤结节后测量肿块的重量、成瘤率 ;彩色多普勒能量图 (CDE)检测转移瘤内新生血管的数目和分布 ;SLAB法免疫组化染色检测转移瘤中微血管密度 (MCD)。结果 局部注射ASPODN组 ,裸鼠胆管癌瘤重及MCD平均值分别为 (2 6.90± 8.4 2 )g和 0 .68± 0 .16,PBS对照组为 (49.3 6± 12 .3 8) g和 2 6.90±8.4 2 (P <0 .0 1) ;局部注射ASPODN组 ,转移瘤内用CDE动态检测到的血管数目及血流丰富程度明显少于PBS组 (P <0 .0 1)。CDE丰富程度与对应切片上MCD的分布基本相符并且成正相关 (P <0 .0 5 )。结论 VEGFASPODN有明显抑制裸鼠胆管癌血管生成和生长的作用。
Objective To observe the inhibitory effect of vascularendothelial growth factor (VEGF) antisense oligodeoxynucleotide (ASPODN) on the angiogenesis and growth of human cholangiocarcinoma in nude mice. Methods The subcultured cholangiocarcinoma cell line QBC93 9 was inoculated subcutaneously in the back of the neck of nude mice at 5 × 10 6 cells / ml. ASPODN and equal amount of PBS were injected into the nude mice from 48 h later. The weight of the tumor was measured after touching the tumor nodules , And tumor formation rate. The number and distribution of neovascularization in metastatic tumor were detected by color Doppler energy imaging (CDE). MAB was detected by SLAB immunohistochemical staining. Results The average tumor weight and MCD in nude mice with local injection of ASPODN were (6.969 ± 8.42) g and 0.68 ± 0.16, respectively, and those in PBS control group were (49.36 ± 12.38) g and 2 6.90 ± 8.42 (P <0.01). In the local injection of ASPODN group, the number of blood vessels and the abundance of blood flow detected by CDE in metastatic tumors were significantly less than those in PBS group (P <0.01). The abundance of CDE basically corresponded positively with the distribution of MCD on the corresponding sections (P <0.05). Conclusion VEGFASPODN can significantly inhibit the angiogenesis and growth of human biliary duct carcinoma in nude mice.
