目的:建立HPLC法同时测定氯甲醑中氯霉素及甲硝唑的含量。方法:采用HPLC法,色谱柱为Nov-pak C18(150mm×4.6 mm,5μm),流动相为甲醇∶水(75∶25),流速为1.0 ml·min-1,检测波长为297 nm,柱温:30℃,进样量20μl。结果:甲硝唑和氯霉素线性范围分别为5~80μg·ml-1(r=0.999 7)和50~800μg·ml-1(r=0.999 8);平均回收率分别为100.83%(RSD=1.82%,n=6)和100.20%(RSD=0.55%,n=6)。结论:本法快速方便、精密度高、重复性和稳定性好,可用于同时测定氯甲醑中氯霉素和甲硝唑的含量。
Objective: To establish an HPLC method for the simultaneous determination of chloramphenicol and metronidazole in chloromethylurea. METHODS: The HPLC method was Nov-Pak C18 (150 mm × 4.6 mm, 5 μm). The mobile phase consisted of methanol and water (75:25) at a flow rate of 1.0 ml · min-1 and the detection wavelength was 297 nm. Temperature: 30 ℃, injection volume 20μl. Results: The linear range of metronidazole and chloramphenicol were 5 ~ 80μg · ml -1 (r = 0.999 7) and 50 ~ 800μg · ml -1 (r = 0.999 8) respectively. The average recoveries were 100.83% (RSD = 1.82%, n = 6) and 100.20% (RSD = 0.55%, n = 6). Conclusion: This method is rapid and convenient, high precision, good repeatability and stability, and can be used for simultaneous determination of chloramphenicol and metronidazole in chloromethylurea.
