目的对局部细胞中HPV16基因表达量及突变与宫颈持续感染的关系进行研究。方法随机选取100例HPV16感染患者,根据3个月复查结果,分成观察组30例和对照组70例,对比观察两组HPV16 E5、E6、E7、LCR基因表达量及突变。结果两组HPV16 E5基因表达量、3978A→C(144L)位点突变以及4041A→G(165V)位点突变对比差异无统计学意义(P>0.05),两组4076A→T(同义突变)位点突变对比差异显著(P<0.05);两组E6、E7、LCR基因表达量及647→G(N29S)联合946T→C(同义突变)位点突变、7761C→T位点突变对比差异无统计学意义(P>0.05),两组178T→G(D25E)位点突变、7727A→C位点突变对比显著(P<0.05)。结论 1局部细胞中HPV16基因表达量与宫颈持续感染无显著关系;2HPV16 E5、E6、E7、LCR基因突变与宫颈持续感染存在相关性,其中HPV16 E5 4076A→T(同义突变)、E6 178T→G(D25E)以及LCR 7727A→C可作为HPV16持续感染的重要突变位点。
Objective To investigate the relationship between HPV16 gene expression in local cells and persistent cervical infection. Methods 100 cases of HPV16 infection were randomly selected and divided into observation group (30 cases) and control group (70 cases) according to the results of 3 months review. The expression and mutation of HPV16 E5, E6, E7 and LCR genes in two groups were compared. Results There were no significant differences in HPV16 E5 gene expression, 3978A → C (144L) site mutation and 4041A → G (165V) site mutation between the two groups (P> 0.05). The 4076A → T (synonymous mutations) (P <0.05). The expression of E6, E7 and LCR genes and the mutation of 647 → G (N29S) combined with 946T → C (synonymous mutation) site and the difference of 7761C → T site mutation in two groups were significantly different There was no significant difference between the two groups (P> 0.05). Mutations of 178T → G (D25E) and 7727A → C in two groups were significant (P <0.05). Conclusions1 HPV16 gene expression in local cells has no significant relationship with persistent cervical infection. There is a correlation between HPV16 E5, E6, E7 and LCR gene mutations and persistent cervical infection. HPV16 E5 4076A → T (synonymous mutation), E6 178T → G (D25E) and LCR 7727A → C are important mutation sites for HPV16 infection.
