目的观察紫茎泽兰提取物对小鼠脾细胞凋亡的影响。方法小鼠分别以0、75、150、300 mg/kg的剂量经口给予紫茎泽兰提取物,连续7 d后,无菌条件下取小鼠脾脏,制备脾细胞悬液,在RPMI 1640培养基中培养24~48 h,流式细胞术观察脾细胞凋亡发生率,光镜下观察脾脏组织病理学变化。结果紫茎泽兰提取物各剂量组脾细胞早期凋亡率、晚期凋亡率及死亡率都明显增加。脾脏组织病理学检查可见,红白髓界限不清,白髓减少或消失,淋巴小结结构破坏,淋巴细胞数量较少,排列疏松,皮质层变薄。结论紫茎泽兰提取物可引起脾脏组织病理学明显改变,脾细胞凋亡率明显增加。
Objective To observe the effect of extract of Eupatorium adenophorum on apoptosis of mouse spleen cells. Methods The mice were orally administered Eupatorium adenophorum extract at doses of 0, 75, 150 and 300 mg / kg, respectively. After 7 days, the spleens of mice were aseptically selected to prepare splenocyte suspension. The cells were cultured in RPMI 1640 The culture medium was cultured for 24-48 h. The incidence of splenocyte apoptosis was observed by flow cytometry. The histopathological changes of spleen were observed under light microscope. Results The early apoptotic rate, the late apoptosis rate and the mortality rate of spleen cells in each dose of Eupatorium adenophorum extract were significantly increased. Histopathological examination of spleen shows that the boundary of red and white marrow is unclear, the white marrow is reduced or disappeared, the structure of lymph node is destroyed, the number of lymphocytes is small, the arrangement is loose and the cortical layer is thinned. Conclusion Eupatorium adenophorum extract can cause significant changes in the histopathology of spleen, and the apoptotic rate of splenocytes is obviously increased.
