目的研究新一代青蒿素抗疟药物ARTPQ重复给药对雌性大鼠生殖内分泌的影响。方法 Wistar大鼠随机分4组,对照组和ARTPQ低、中、高剂量组(35、70、120 mg/kg),每组15只,ig给药14 d。给药后每天测量大鼠动情周期,给药结束后采集大鼠血液,Elisa方法检测大鼠血液雌激素、肾上腺皮质激素水平,肉眼观察各脏器变化及称量大鼠子宫、卵巢、肾脏及肾上腺的质量。结果连续给药3 d后大鼠动情周期紊乱(维持于动情后期或间期),大鼠子宫未见有扩张,平均质量低于对照组。ARTPQ低、中、高剂量组大鼠肾上腺系数高于对照组(P<0.05、0.01)。ARTPQ高剂量组大鼠雌激素水平低于对照组(P<0.05),而ARTPQ低、中、高剂量组大鼠肾上腺素水平高于对照组(P<0.05、0.01)。结论 ARTPQ作用于肾上腺,肾上腺增生,肾上腺皮质激素的水平升高,雌激素水平降低,表现为大鼠发情周期的紊乱。
Objective To study the effects of a new generation of artemisinin antimalarial drug ARTPQ on reproductive endocrine in female rats. Methods Wistar rats were randomly divided into 4 groups, control group and ARTPQ low, medium and high dose groups (35, 70, 120 mg / kg), 15 in each group and 14 d in ig group. The estrous cycle of rats was measured daily after administration, the blood of rats was collected after administration, the levels of blood estrogen and adrenocorticotropic hormone in rats were measured by Elisa method, the changes of various organs were observed with naked eye, and the uterus, ovaries, kidneys and Adrenal quality. Results After 3 days of continuous administration, the estrous cycle of rats was disturbed (maintained at late estrus or interphase). No expansion was seen in the uterus of rats, and the average quality was lower than that of the control group. The ARTPQ low, middle and high dose group rats adrenal coefficient higher than the control group (P <0.05,0.01). The level of estrogen in ARTPQ high dose group was lower than that in control group (P <0.05), while the level of epinephrine in ARTPQ low dose group was higher than that in control group (P <0.05, 0.01). Conclusion ARTPQ acts on the adrenal glands, adrenal hyperplasia, adrenocorticotropic hormone levels increased, estrogen levels decreased, the performance of the rat estrous cycle disorders.
