采用大孔树脂SP825分离红霉素A(EA)及其主要杂质红霉素C(EC)。通过平衡试验考察了溶剂极性对SP825吸附EA和EC的影响;以洗涤法和迎头色谱法分离EA和EC,考察洗涤流速、洗涤液pH值对洗涤曲线的影响,并对比了两种分离方式的洗脱产品。结果表明,含3%乙酸乙酯的磷酸盐缓冲液为极性最佳的溶剂;洗涤流速10.5 ml/h、pH 7.0较适宜;洗涤法与迎头色谱法均能实现EA与EC的有效分离,综合考虑成本及分离周期,迎头色谱法优于洗涤法。
The macroporous resin SP825 was used to separate erythromycin A (EA) and its major impurity erythromycin C (EC). The effects of solvent polarity on the adsorption of EA and EC on SP825 were investigated by the equilibrium test. The EA and EC were separated by washing and on-line chromatography. The effects of washing flow rate and washing solution pH on the washing curve were investigated. Of the elution product. The results showed that the best polar solvent was phosphate buffer solution containing 3% ethyl acetate. The washing flow rate was 10.5 ml / h and pH 7.0 was suitable.
