目的观察帕罗西汀对心理应激大鼠海马5-HT表达的影响,以揭示帕罗西汀治疗心理应激引起焦虑的中枢作用机制。方法建立大鼠心理应激模型,观察各组大鼠血浆皮质醇、旷场行为和海马5-HT的表达。结果与正常对照组比较,各应激组大鼠血浆皮质醇含量明显升高,旷场活动明显增多,海马5-HT表达减少,而帕罗西汀给药能减少应激大鼠血浆皮质醇含量和旷场活动,增加5-HT表达。结论帕罗西汀可能是通过增加海马5-HT表达,保护了海马的功能,下调HPA轴水平,从而发挥了中枢抗焦虑作用。
Objective To observe the effect of paroxetine on 5-HT expression in hippocampus of psychological stress rats to reveal the central mechanism of paroxetine in the treatment of psychological stress-induced anxiety. Methods The rat model of psychological stress was established. The plasma cortisol, open field behavior and the expression of 5-HT in hippocampus of rats in each group were observed. Results Compared with the normal control group, the plasma cortisol levels of rats in each stress group were significantly increased, the open field activity was significantly increased, and the expression of 5-HT was decreased in hippocampus. Paroxetine administration could reduce the plasma cortisol level and Open field activity increases 5-HT expression. Conclusion Paroxetine may play an central role in anti-anxiety by increasing the expression of 5-HT in the hippocampus, protecting the function of the hippocampus and lowering the HPA axis.
