采用单次腹腔注射野百合碱(MCT)的方法复制大鼠肺心病模型,芪参益气滴丸浸膏灌胃给药6周后,通过HPLC检测大鼠右心肌组织中的腺苷酸(ATP,ADP,AMP)的含量,进而考察芪参益气滴丸对肺心病大鼠心肌能量代谢的影响。结果显示ATP,ADP,AMP达到良好分离;ATP,ADP,AMP在考察的浓度范围内,线性关系良好;加样回收率均在90%~108%;与模型组相比,高剂量给药组大鼠心肌中ATP的含量显著性升高,ADP的含量有升高趋势,AMP含量有下降趋势。提示芪参益气滴丸能够改善心肌组织能量代谢系统。通过HPLC,为测定心肌组织中ATP,ADP,AMP含量提供了一种快速、高效的定性定量分析方法,为生物样品中能量代谢相关含量的测定研究提供了参考。
A single intraperitoneal injection of monocrotaline (MCT) was used to replicate the model of pulmonary heart disease in rats. Six weeks after the administration of Qishenyiqi drip pills, the adenosine monophosphate ATP, ADP and AMP), and then investigate the Qishenyiqi dripping pills on myocardial energy metabolism in rats with cor pulmonale. The results showed that ATP, ADP and AMP reached a good separation. ATP, ADP and AMP showed good linearity within the investigated concentration range. The recoveries were all within the range of 90% -108%. Compared with the model group, The content of ATP in rat myocardium increased significantly, the content of ADP increased and the content of AMP decreased. It suggests that Qishenyiqi Dripping Pills can improve myocardial energy metabolism system. HPLC provides a rapid and efficient qualitative and quantitative analysis method for the determination of ATP, ADP and AMP in myocardial tissue, which provides a reference for the determination of the related content of energy metabolism in biological samples.
