目的:观察血管紧张素Ⅱ(AngⅡ)对培养乳鼠心肌细胞多聚二磷酸腺苷(ADP)核糖合成酶(PARP)活性和表达的影响。方法:新生大鼠心肌原代培养,用10-7mol/LAngⅡ诱导,观察心肌细胞活性氧(ROS)的生成、PARP活性与PARP1蛋白表达情况,并观察缬沙坦与维生素C预处理对AngⅡ诱导心肌细胞ROS的生成、PARP活性与PARP1蛋白表达的影响。结果:AngⅡ诱导心肌细胞ROS生成增加,心肌细胞PARP激活,PARP1蛋白表达增加。给予缬沙坦与维生素C预处理后心肌细胞ROS生成减少,PARP活性降低,PARP1蛋白表达减少。结论:AngⅡ可以诱导培养的心肌细胞ROS生成增加,PARP激活,PARP1蛋白表达增加,PARP激活ROS可能是AngⅡ诱导心肌重构机制之一。
Objective: To observe the effect of Ang Ⅱ on the activity and expression of poly ADP ribose synthase (PARP) in neonatal rat cardiomyocytes. Methods: Primary cultured neonatal rat cardiomyocytes were induced with 10-7mol / LAngⅡ to observe the generation of reactive oxygen species (ROS), the activity of PARP and the expression of PARP1 in cardiomyocytes. The effects of valsartan and vitamin C preconditioning on the induction of AngⅡ Generation of ROS in cardiomyocytes, PARP activity and PARP1 protein expression. Results: AngⅡinduced ROS generation in cardiomyocytes, activation of PARP in cardiomyocytes and increase of PARP1 protein expression. After pretreatment with valsartan and vitamin C, ROS production decreased, PARP activity decreased and PARP1 protein expression decreased. CONCLUSION: AngⅡ can induce the ROS production in cultured cardiomyocytes, and increase PARP activation and PARP1 protein expression. PARP-activated ROS may be one of the mechanisms of AngⅡ-induced myocardial remodeling.
