目的从四种蛋白酶中选出酶解全蝎粉的最佳用酶,来制备抗肿瘤多肽。方法以蛋白质的水解度、对肺腺癌细胞A549细胞的抑制率为指标,同时以全蝎粉水提物作为对照组,评价胃蛋白酶、胰蛋白酶、胰凝乳蛋白酶、弹性蛋白酶对全蝎粉的水解效果。结果胃蛋白酶对全蝎粉的水解度为8.47%,且不同的药物浓度对肺腺癌A549细胞的抑制率比对照组低,其IC50为14.7427 mg/ml;胰蛋白酶、胰凝乳蛋白酶、弹性蛋白酶的水解度分别为25.12%、12.37%、11.13%,不同的药物浓度对肺腺癌A549细胞的抑制率比对照组高,其IC50分别为0.6878 mg/ml、0.7073 mg/ml、0.8306 mg/ml;对照组的IC50为5.9386 mg/ml。结论全蝎粉酶解工艺的最佳用酶是胰蛋白酶,其次是胰凝乳蛋白酶和弹性蛋白酶,而胃蛋白酶可以去除。
