目的:建立一测多评法测定苦豆子总生物碱中6种成分的含量。方法:采用HPLC法,DIKMA C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈(A)-0.05 mol/L KH2PO4溶液(含2 mL/L三乙胺)(B),梯度洗脱;柱温:30℃;流速:1.0 mL/min;检测波长:205 nm。以槐定碱作为参照物,建立金花雀碱、槐果碱、苦参碱、氧化苦参碱及莱曼碱的相对校正因子,分别采用一测多评法和外标法测定苦豆子总生物碱中6种成分的含量,评价一测多评法计算值与外标法实测值的差异。结果:一测多评法计算值与外标法实测值之间没有显著性差异,所得的相对校正因子可信。结论:一测多评法可用于苦豆子总生物碱的质量评价,方法可靠、准确。
Objective: To establish a test to determine the content of six components in total alkaloids of Sophora alopecuroides. Methods: The mobile phase consisted of acetonitrile (A) - 0.05 mol / L KH2PO4 solution (containing 2 mL / L triethylamine) (B) on a DIKMA C 18 column (250 mm × 4.6 mm, , Gradient elution; column temperature: 30 ℃; flow rate: 1.0 mL / min; detection wavelength: 205 nm. Using sophoridine as a reference, the relative correction factors of goldenseal, sophocarpine, matrine, oxymatrine and Lemanine were established, respectively. Alkaloids in the six components of the content, evaluation of a measured multiple evaluation of the calculated value and external standard measured difference. Results: There is no significant difference between the measured value of multiple evaluation method and the actual value of external standard method, and the relative correction factor obtained is credible. Conclusion: A multiple assessment method can be used for the quality evaluation of total alkaloids from Sophora alopecuroides, the method is reliable and accurate.
