目的 观察脑梗死患者循环白细胞对培养内皮细胞金属蛋白酶 (MMP) 9表达的影响 ,探讨循环白细胞及内皮细胞在脑梗死病理机制中的作用。方法 采用细胞培养及免疫荧光技术 ,分别将 6例脑梗死患者及 6例正常人循环白细胞与内皮细胞联合培养 ,激光共聚焦成像定量测定内皮细胞MMP 9的表达水平。结果 与脑梗死患者循环白细胞联合培养的内皮细胞均荧光着色 ,显示MMP 9中度~强度表达 ,表达部位主要在胞浆 ,而与正常人白细胞联合培养的内皮细胞则基本无荧光着色 ;激光共聚焦成像显示脑梗死患者组荧光强度值为 131± 34,正常人组为 2 0± 8,两组比较P <0 0 0 1,差异有显著意义。结论 提示脑梗死患者循环白细胞处于炎性状态 ,可刺激内皮细胞表达MMP 9,并可能由此导致血浆MMP 9水平增高 ,抑制循环白细胞活性及血管内皮细胞MMP 9的表达 ,应成为脑梗死防治的重要有效途径之一。
Objective To investigate the effect of circulating leukocytes on the expression of metalloproteinase (MMP) 9 in cultured human cerebral infarction patients and to explore the role of circulating leukocytes and endothelial cells in the pathogenesis of cerebral infarction. Methods Cell culture and immunofluorescence were used to detect the expression of MMP-9 in 6 cases of cerebral infarction and 6 cases of normal human circulatory leukocytes and endothelial cells respectively. Confocal laser scanning microscopy was used to detect the expression of MMP-9. RESULTS: The endothelial cells co-cultured with circulating white blood cells in patients with cerebral infarction were all fluorescently stained, showing moderate to strong expression of MMP-9, mainly in the cytoplasm, and almost no fluorescence staining in endothelial cells co-cultured with normal white blood cells. Focus imaging showed that the fluorescence intensity of patients with cerebral infarction group was 131 ± 34, normal group was 20 ± 8, the two groups compared P <0 0 0 1, the difference was significant. Conclusions These results suggest that circulating leukocytes are inflammatory in patients with cerebral infarction and may stimulate the expression of MMP-9 in endothelial cells, which may result in the increase of plasma MMP-9, inhibition of circulating leukocyte activity and MMP-9 expression in vascular endothelial cells One of the important and effective ways.
