目的 观察复方鳖甲软肝方 (FFBJRGF)药物血清干预TGF β1对离体肝星形细胞 (HSC)增殖的影响作用。方法 采用改进的中药血清药理学方法 ,在HSC的培养体系中添加外源性TGF β1,运用MTT法检测TGF β1对HSC增殖的影响 ,观测不同剂量复方鳖甲软肝方药物血清的干预TGF β1对离体肝星形细胞 (HSC)增殖的作用。结果 添加外源性TGF β1可减低HSC的增殖能力 ,但显效时间较长(72h) ;高、中、低剂量FFBJRGF药物血清组和IFN -γ血清组与正常对照HSC培养组比较 ,均明显HSC的增殖活性 (各组OD值比较 ,分别P <0 0 5 ) ,且显效时间短于添加外源性TGF β1组(2 4h)。结论 FFBJRGF药物血清、IFN γ血清和TGF β1均具有减弱HSC增殖的作用 ,且FFBJRGF药物血清可在TGF β1作用基础上发挥增效作用
Objective To observe the effect of FFBJRGF drug serum on the proliferation of isolated hepatic stellate cells (HSC) by intervention of TGFβ1. Methods The improved serum pharmacology method of traditional Chinese medicine was used to add exogenous TGFβ1 in the culture system of HSC. The effect of TGFβ1 on the proliferation of HSC was detected by MTT assay. Effect on the proliferation of isolated hepatic stellate cells (HSCs). Results Exogenous TGFβ1 could reduce the proliferation of HSC, but the effective time was longer (72h). The high, middle and low doses of FFBJRGF sera and IFN-γ serogroup were significantly higher than that of the normal control HSC. The proliferative activity (comparison of OD values in each group, respectively, P <0 05), and markedly shorter than the addition of exogenous TGFβ1 group (24 hours). Conclusion FFBJRGF drug serum, IFN γ serum and TGF β1 all have the effect of weakening HSC proliferation, and FFBJRGF drug serum can exert synergism on the basis of TGF β1 effect.
