在单因素试验的基础上,采用Box-Behnken设计对大叶金花草总黄酮纤维素酶提取工艺中的液料比、pH值和酶解温度3因素的最优化组合进行了定量研究,建立并分析了各因素与得率关系的数学模型。结果表明:最佳的工艺条件为液料比10.8mL/g、pH值4.6和酶解温度48℃;经试验验证,在此条件下得率为6.58%,与理论计算值6.64%基本一致,说明回归模型能较好地预测大叶金花草中总黄酮的提取得率。
Based on the single-factor experiment, the optimal combination of three factors, liquid-solid ratio, pH value and enzymolysis temperature, was determined by Box-Behnken design. The mathematical model of the relationship between factors and yield was analyzed. The results showed that the optimal process conditions were liquid-to-liquid ratio of 10.8mL / g, pH value of 4.6 and enzymolysis temperature of 48 ℃. The experimental results showed that the yield was 6.58%, which was basically consistent with the calculated value of 6.64% It shows that the regression model can better predict the extraction yield of total flavonoids in S. australis.
