目的探讨急性髓细胞白血病(AML)患者组蛋白乙酰化修饰规律,并研究组蛋白乙酰化对过氧化还原酶(PRDX2)基因差异表达的调控作用。方法应用实时荧光定量聚合酶链反应(PCR)技术检测55例AML患者和20例非白血病患者单个核细胞(MNC)的PRDX2mRNA的表达,应用蛋白印迹法检测乙酰化H3表达情况。结果 AML组mRNA表达量较对照组明显下降,差异有统计学意义(t=-4.89,P<0.05);2组乙酰化H3相对表达量为2.2±1.0,3.2±0.9,差异有统计学意义(t=-3.94,P<0.05);AML组患者组蛋白H3乙酰化水平与PRDX2基因mRNA呈正相关(r=0.358,P=0.007)。结论 AML患者的组蛋白乙酰化呈低表达现象,组蛋白乙酰化在AML患者中对PRDX2基因差异表达具有调控作用。
Objective To investigate the regulation of histone acetylation in patients with acute myeloid leukemia (AML) and to investigate the role of histone acetylation in the differential expression of peroxiredoxin gene (PRDX2). Methods The expression of PRDX2 mRNA in 55 patients with AML and 20 patients with non-leukemia was detected by real-time fluorescence quantitative polymerase chain reaction (PCR). The expression of acetylated H3 was detected by Western blotting. Results The mRNA expression of AML was significantly lower than that of the control group (t = -4.89, P <0.05). The relative expression of acetylated H3 in the two groups was 2.2 ± 1.0 and 3.2 ± 0.9, the difference was statistically significant (t = -3.94, P <0.05). The histone H3 acetylation level in AML group was positively correlated with PRDX2 mRNA (r = 0.358, P = 0.007). Conclusions The histone acetylation of AML patients is low expression, and histone acetylation regulates the differential expression of PRDX2 gene in AML patients.
