目的:观察CXCR4腺病毒表达载体(adenovirus vector of CXCR4,Adv-CXCR4)转染的真皮多能干细胞(dermal multipotent stem cells,dMSCs)移植后可否更多地向全身照射大鼠骨髓分布,并加速骨髓造血功能恢复。方法:3H-TdR标记Adv-CXCR4转染的dMSCs(A组)、绿色荧光蛋白腺病毒表达载体转染的dMSCs(B组)、未转染病毒载体的dMSCs(C组)后移植到全身照射大鼠体内,液闪法测量骨髓中dMSCs的含量,监测血中红细胞等的恢复情况。结果:伤后5d起,A组骨髓中dMSCs的含量明显高于B组和C组(P<0.05)。伤后14d和21d,A组红细胞、白细胞和血小板的数量明显高于B组和C组(P<0.05)。结论:Adv-CXCR4转染的dMSCs在移植后可更多地分布到全身照射大鼠的骨髓,且能更快地促进骨髓造血功能的恢复。
OBJECTIVE: To investigate whether the transplantation of dermal multipotent stem cells (DMSCs) transfected with the CXCR4 adenovirus vector of CXCR4 can increase the bone marrow distribution to the whole body and accelerate bone marrow Hematopoietic function recovery. Methods: The dMSCs transfected with Ad-CXCR4 by 3H-TdR (group A), dMSCs transfected with GFP expression vector (group B) and dMSCs without retroviral vector (group C) were transplanted into whole body Rats in vivo, liquid scintillation method of measuring bone marrow dMSCs content, blood red blood cells and other recovery monitoring. Results: From the 5th day after injury, the content of dMSCs in group A was significantly higher than those in group B and C (P <0.05). On the 14th and 21st day after injury, the number of erythrocytes, white blood cells and platelets in group A was significantly higher than those in groups B and C (P <0.05). CONCLUSION: Adv-CXCR4-transfected dMSCs can be more distributed to the bone marrow of rats exposed to whole body irradiation and can promote the recovery of hematopoietic function of bone marrow more rapidly.
