目的　检测炎前因子 m RNA在增生性玻璃体视网膜病变 (proliferative vitreoretinopathy,PVR)中的表达情况 ,证实其在 PVR发病中的作用。　方法　收集 14例不同分级的 PVR患者玻璃体切割术中所取出的增生膜 ,石蜡包埋切片 ,用生物素标记的寡核苷酸探针进行原位杂交 ,检测 IL- 1β、IL- 6、IL- 8和 TNF- α m RNA的表达 ,两例角膜移植后的供体眼作为正常对照。　结果　共有 5例样本有阳性表达。 3例增生膜中 IL- 1βm RNA表达 ,3例表达了 IL- 6 m RNA,1例表达了 IL- 8m RNA,3例表达 TNF- α m RNA。其中 1例既表达了 IL- 1β又表达了 IL- 6 m RNA,1例 IL- 1β、IL- 8和 TNF- α m RNA三者均表达 ,2例有 IL- 6和 TNF- α m RNA两者表达。正常视网膜未检测到炎前因子 m RNA的表达。　结论　IL- 1β、IL- 6、IL- 8和TNF- α参予了 PVR增生膜的形成过程。 Objective To detect the expression of proinflammatory cytokine m RNA in proliferative vitreoretinopathy (PVR) and confirm its role in the pathogenesis of PVR. Methods Fourteen PVR patients with different grading were collected for proliferative membrane and paraffin embedded sections during vitrectomy and biotin-labeled oligonucleotide probes were used for in situ hybridization. The levels of IL-1β, IL-6, IL - 8 and TNF-α m RNA expression, two cases of corneal donor eye as a normal control. Results A total of 5 samples were positive. IL-1βm RNA was expressed in 3 cases of proliferative membrane, IL-6m RNA was expressed in 3 cases, IL-8m RNA was expressed in 1 case, and TNF-αm RNA was expressed in 3 cases. One of them expressed both IL-1β and IL-6 m RNA, one of them expressed IL-1β, IL-8 and TNF-αm RNA, and two of them expressed IL-6 and TNF-αmRNA Both expressed. Normal retina did not detect the expression of pre-inflammatory factor m RNA. Conclusion IL-1β, IL-6, IL-8 and TNF-α participate in the process of PVR hyperplasia.
