实验观察了NG108-15和SK-N-SH细胞在含川楝素的培养基中产生的形态学变化。(1)光镜下发现,川楝素可以引起NG108-15细胞突起变短甚至消失、胞体变圆、细胞萎缩、失去贴壁能力出现飘浮,数量逐渐减少;在有血清培养的情况下,川楝素引起SK-N-SH细胞间界限不清,无血清时,细胞出现飘浮现象。(2)电镜观察显示,川楝素引起NG108-15细胞和SK-N-SH细胞的死亡,大多数具有坏死细胞的特征,仅个别细胞的死亡具有典型凋亡细胞的特征。
Morphological changes of NG108-15 and SK-N-SH cells in toosendanin-containing medium were observed experimentally. (1) It was found by light microscope that toosendanin could cause the protrusion of NG108-15 cells to shorten or even disappear, the cell body became round, the cell atrophy, the ability to lose the adhesion appeared to float and the number decreased gradually; in the case of serum culture, Toosendanin caused the unclear boundaries between SK-N-SH cells. When serum-free, the cells appeared floating. (2) Electron microscopy showed that toosendanin caused the death of NG108-15 cells and SK-N-SH cells, most of which had the characteristics of necrotic cells. The death of only individual cells was characterized by typical apoptotic cells.
