本研究旨在阐明对造血调控中起重要作用的Ikaros家族成员Ikaros和Helios的两个异构体Ik-6和He-i的表达特征、发生机制、临床意义和关联性。对163例白血病患者的骨髓细胞进行了PCR分析。结果显示,Ik-6和He-i高表达于不同的白血病亚型,Ik-6高表达于BCR/ABL融合基因阳性的急性淋巴细胞白血病,而He-i则高表达于T细胞急性淋巴细胞白血病。克隆和测序结果显示,异构体Ik-6和He-i具有不同的遗传学改变。本研究未观察到Ik-6和He-i之间具有统计学意义的关联性。结论:虽然Ikaros和Helios是具有极其相似锌指结构的同一家族成员,但其异构体Ik-6和He-i在白血病中的表达特征、发生机制和临床意义有差异。Ik-6和He-i缺失的外显子分别含有或不含有DNA结合功能域,这种差异可能导致两者具有不同的生物学功能。两者之间的相互作用和关联性有待深入研究。
The aim of this study was to elucidate the molecular mechanisms of expression, pathogenesis, clinical significance and relevance of the two isoforms Ik-6 and He-i of Ikaros and Helios, members of the Ikaros family that play an important role in the regulation of hematopoiesis. The bone marrow cells from 163 patients with leukemia were analyzed by PCR. The results showed that Ik-6 and He-i were highly expressed in different leukemia subtypes, Ik-6 was highly expressed in BCR / ABL fusion gene-positive acute lymphoblastic leukemia, while He-i was highly expressed in T lymphocyte acute lymphocytes leukemia. The cloning and sequencing results showed that the isoforms Ik-6 and He-i have different genetic changes. No statistically significant associations between Ik-6 and He-i were observed in this study. CONCLUSION: Although Ikaros and Helios are members of the same family with extremely similar zinc finger motifs, the expression patterns, pathogenesis, and clinical significance of their isoforms Ik-6 and He-i in leukemia are different. The deletion of Ik-6 and He-i exons, respectively, with or without DNA binding domain, may lead to different biological functions between the two. The interaction and relevance between the two needs further study.
