目的 观察不同胎龄大鼠小脑提取液 (cerebellum extracts,CE)对体外培养小脑神经细胞的影响。方法 制备不同蛋白浓度的 14、16、18、2 0 d胎鼠 (E14、E16、E18、E2 0 )及新生 1d鼠 (P1) CE,加入原代培养的E18胎鼠小脑神经细胞中 ,2 4h后以微量快速自动比色法检测细胞活性。结果 实验组各胎龄 CE均能促进培养小脑神经元的存活 ,并有蛋白浓度依赖关系 ,各胎龄都有各自不同的最适浓度 ,E16和 E18CE显示了更强的神经营养活性 ,与对照组及其他实验组相比差异有极显著性 (P<0 .0 0 1)。结论 CE对培养小脑细胞具有高度神经营养活性。
Objective To observe the effect of cerebrellum extracts (CE) of different gestational age rats on cultured cerebellar neurons. Methods E14, E16, E18, E2 0, and embryonic day 1d (P1) CE were cultured in 14,16,18 and 20 day embryos with different protein concentrations. Primary cultured E18 fetal rat cerebellar neurons were cultured. After 4h, the cell viability was detected by micro-rapid automated colorimetry. Results All gestational age groups in experimental group were able to promote the survival of cultured cerebellar neurons, and there was protein concentration-dependent relationship. Each of the gestational ages had different optimal concentrations. E16 and E18CE showed stronger neurotrophic activity compared with the control There was significant difference between the experimental group and other experimental groups (P <0.01). Conclusion CE has high neurotrophic activity on cultured cerebellar cells.
