目的:建立金莲花的薄层鉴别及其总黄酮含量的测定方法,为完善该药材的质量控制体系提供参考。方法:通过单因素试验筛选提取溶剂和展开系统,建立金莲花的TLC。选择荭草苷为指标成分,采用比色法测定总黄酮含量,以10%硝酸铝溶液为显色剂,检测波长510 nm,考察不同产地金莲花中总黄酮含量的差异。结果:展开剂为三氯甲烷-甲醇-丁酮-冰乙酸(4∶2∶3∶3),建立的TLC分离效果良好。荭草苷线性范围8.096~56.672 mg·L-1,平均加样回收率为98.8%;1,2,3,4,5,6号样中总黄酮质量分数分别为9.79%,8.71%,8.32%,6.47%,6.42%,6.71%。结论:建立的金莲花定性鉴别和定量检测方法简便、准确,适用于该药材的质量控制。
OBJECTIVE: To establish a method for TLC identification and determination of total flavonoids content of Nasturtium officinale, and provide reference for improving the quality control system of this medicinal material. Methods: Single factor experiments screening solvent extraction and development system, the establishment of the Trollius TLC. Tollucoside was selected as the index component, and the content of total flavonoids was determined by colorimetric method. The detection wavelength was 510 nm with 10% aluminum nitrate solution as the chromogenic reagent. Results: The developing solvent was chloroform-methanol-butanone-glacial acetic acid (4: 2: 3: 3) The linear range of hesperidin was 8.096 ~ 56.672 mg · L-1, the average recovery rate was 98.8%. The total flavonoids contents of 1,2,3,4,5,6 were 9.79%, 8.71%, 8.32 %, 6.47%, 6.42%, 6.71%. Conclusion: The established method of qualitative identification and quantitative detection of Trollius niloticus is simple and accurate, which is suitable for the quality control of this herb.
