目的研究五加皮提取成分(Acanthopanaxgracilistylusextract,Age)对肿瘤细胞的作用机理。方法采用〔3H〕TdR掺入法测定肿瘤细胞增殖反应;用流式细胞技术分析Age对肿瘤细胞周期的影响;用蛋白印迹技术检测Age对肿瘤细胞Rb(retinoblastoma)、Cdk(cyclindependentkinases)等酶类的影响。结果体外细胞活力试验证明,Age仅抑制肿瘤细胞增殖,并不导致细胞死亡。在Age的作用下,肿瘤细胞增殖停止在细胞周期的G0/G1期,而未见直接的细胞毒效应。Age可诱导Rb、Cdk2和Cdk4表达降低,使细胞停止增殖。结论Age对肿瘤细胞的作用机制是通过调节控制细胞周期的酶类而发挥作用的。
Objective To study the mechanism of action of Acanthopanax gracilistyl usextract (Age) on tumor cells. Methods The proliferation of tumor cells was measured by [3H]TdR incorporation method; the effect of Age on tumor cell cycle was analyzed by flow cytometry; Age was used to detect the enzyme Rb (retinoblastoma), Cdk (cyclindependent kinases) and other enzymes. Impact. Results In vitro cell viability assays demonstrated that Age inhibited tumor cell proliferation only and did not lead to cell death. Under the action of Age, tumor cell proliferation stopped at the G0/G1 phase of the cell cycle, and no direct cytotoxic effect was observed. Age induces a decrease in the expression of Rb, Cdk2, and Cdk4 and stops cells from proliferating. Conclusion The action mechanism of Age on tumor cells plays a role by regulating the enzymes that control the cell cycle.
