该实验为探究覆盆子乙酸乙酯提取物对去卵巢阿尔茨海默病(AD)模型小鼠海马蛋白组表达的影响。小鼠去卵巢后按体重随机分成假手术组、模型组、阳性药对照组(雌二醇,0.1 g·L~(-1))和覆盆子乙酸乙酯部位组(生药浓度18 g·kg~(-1))。阳性对照组小鼠隔天皮下注射雌二醇0.2 m L/只,覆盆子乙酸乙酯部位组灌胃覆盆子乙酸乙酯提取物0.01 m L·g~(-1),模型组与假手术组灌胃生理盐水0.01 m L·g~(-1),每日1次,连续给药32 d。分离小鼠海马,提取蛋白,Nanol-ESI的液相-质谱联用系统检测,Protein Discovery软件进行蛋白鉴定,SIEVE软件对海马蛋白进行相对定量定性分析。实验结果显示,覆盆子乙酸乙酯组与模型组相比较共有66个差异表达蛋白,包含热休克蛋白、微管蛋白、能量代谢相关蛋白和脑保护相关蛋白等与AD相关的蛋白。以上差异蛋白可能是覆盆子有效部位防治AD的靶点蛋白。
The experiment was conducted to investigate the effect of the ethyl acetate extract of raspberry on the protein expression of hippocampus in ovariectomized Alzheimer’s disease (AD) model mice. After ovariectomized, the mice were randomly divided into sham operation group, model group, positive control group (estradiol, 0.1 g · L -1) and ethyl acetate raspberry group (crude drug concentration 18 g · kg ~ (-1)). Mice in the positive control group were injected subcutaneously with estradiol (0.2 m L) every other day, while those in the raspberry group were orally administered with raspberry ethyl acetate extract 0.01 m L · g -1. The model group and sham operation Groups were given normal saline 0.01 m L · g -1 once daily for 32 days. Separation of mouse hippocampus, protein extraction, Nanol-ESI liquid-mass spectrometry system, Protein Discovery software for protein identification, SIEVE software for quantitative and qualitative analysis of hippocampal proteins. The results showed that compared with the model group, there were 66 differentially expressed proteins in the ethyl acetate raspberry group, which were related to AD, including heat shock proteins, tubulin, energy metabolism related proteins and brain protection related proteins. The above differential proteins may be the active site of raspberry prevention and treatment of AD target protein.
