有关油菜的组织培养已有许多报道,但多局限于采用不同外植体获得再生植株,对于影响外植体高频率再生的诸因素未作系统分析。在油菜的基因转化中外植体的来源和再生方式都直接影响着转化效果。根据Mukhopadlyay(1992)等人和Radke(1992)的工作,下胚轴是较好的外植体来源,而直接成苗的再生方式最大限度地保存了原种质的遗传特性。我们对芥菜型油菜下胚轴高频率植株再生的因素进行了研究,以期为油菜基因转化工作中高效外植体再生系统的建立提供依据。
Tissue culture of rapeseed has been reported in many cases, but more limited to the use of different explants to obtain regenerated plants, the factors affecting the high frequency of explant regeneration without systematic analysis. In the gene transformation of rape, the origin and regeneration of explants all have a direct impact on transformation efficiency. Hypocotyls are a good source of explants according to the work of Mukhopadlyay (1992) et al. And Radke (1992), whereas direct seedling regeneration maximizes the inheritance of the original germplasm. We studied the regeneration of high-frequency plants in Brassica juncea hypocotyls in order to provide a basis for the establishment of efficient explants regeneration system in the gene transformation of rapeseed.
