目的:观察细胞外信号调节激酶(ERK)、Wnt/β-连环蛋白(β-catenin)信号通路相关蛋白在右归饮含药血清影响成骨细胞过程中的作用,明确右归饮含药血清对ERK、Wnt通路和ERK/Wnt交叉串扰的作用。方法:体外培养成骨细胞,分组,加药干预。培养3d后,应用ELISA检测Wnt/β-catenin及ERK通路相关蛋白β-catenin、Runx2、ERK蛋白表达情况。结果:右归饮含药血清能显著上调3种蛋白表达(P<0.05,P<0.01)。联合阻断ERK、Wnt通路,3种蛋白表达水平较含药血清组有明显降低(P<0.05,P<0.01);与单独阻断其中一条通路比较,有明显下降(P<0.01)。单独阻断某一条通路,仅Runx2较右归饮含药血清组有明显降低(P<0.05,P<0.01)。结论:右归饮含药血清能显著增加成骨细胞活性,该作用与ERK、Wnt信号通路密切相关,且两条通路之间存在一定的交叉串扰作用。
OBJECTIVE: To observe the role of ERK and Wnt / β-catenin signaling pathway-related proteins in the osteoblasts affected by Yougui Yin-containing serum, Effect on ERK, Wnt pathway and ERK / Wnt cross crosstalk. Methods: Osteoblasts were cultured in vitro and divided into groups and medicated. After cultured for 3 days, the protein expressions of Wnt / β-catenin and ERK pathway-related proteins β-catenin, Runx2 and ERK were detected by ELISA. Results: Youguiyin medicated serum could significantly up-regulate the expression of three proteins (P <0.05, P <0.01). Compared with the drug-containing serum, the levels of ERK and Wnt pathway were significantly decreased (P <0.05, P <0.01). Compared with blocking one of them, the expression of ERK and Wnt was significantly decreased (P <0.01). Only blocking a single pathway, Runx2 than the right to drink Yin containing serum group was significantly lower (P <0.05, P <0.01). Conclusion: Youguiyin containing serum can significantly increase the activity of osteoblasts, which is closely related to ERK and Wnt signaling pathways, and there is a certain crosstalk between the two pathways.
