目的:优选中药麦芽发芽炮制工艺。方法:以麦芽中总生物碱含量、大麦芽碱含量及淀粉酶活力为指标,在单因素试验基础上,通过正交试验考察浸泡时间、发芽温度、发芽湿度及每日洒水量对大麦发芽的影响。依次采用酸性染料比色法和高效液相色谱法测定总生物碱与大麦芽碱含量,DNS比色法测定淀粉酶活力。结果:优选出的麦芽最佳发芽工艺为:用水浸泡5h,发芽温度25℃,湿度70%,每日洒水量与大麦重量比为1∶1。结论:优选得到的炮制工艺稳定可行。
Objective: To optimize the malt germination of traditional Chinese medicine processing technology. Methods: Based on single factor experiments, the total alkaloid content, malt alkaloid content and amylase activity in malt were used as indexes to study the effects of soaking time, germination temperature, germination humidity and daily watering on germination of barley influences. The content of total alkaloids and barley malt alkaloids were determined by acid dye colorimetry and high performance liquid chromatography respectively. The amylase activity was determined by DNS colorimetry. Results: The optimal germination process of malt was as follows: water soaked for 5h, germination temperature 25 ℃, humidity 70%, daily watering and barley weight ratio 1: 1. Conclusion: The optimized processing technology is stable and feasible.
