目的 研究小檗碱对大鼠肝脏基因表达谱的影响。方法 大鼠予以盐酸小檗碱灌胃 6d ,摘取肝脏提取总RNA。用Cy3和Cy5两种荧光物质分别标记阴性对照组和盐酸小檗碱组肝组织cDNA ,制备成探针与表达谱芯片进行杂交 ,获得的荧光信号图像用计算机扫描后进行数据分析 ,计算每点的Cy5和Cy3信号强度比值。结果 在被检测的 4 0 96个基因中 ,盐酸小檗碱组差异表达基因 2 36个 ,与代谢相关的差异表达基因共 32个 ,其中表达上调的代谢相关基因 8个 ,包括大鼠环核苷酸磷酸二酯酶、异柠檬酸脱氢酶 3、心肌黄酶和葡萄糖 6 磷酸酶等。表达下调的代谢相关基因 2 4个 ,包括尿苷二磷酸 葡糖醛酰转移酶、细胞色素P4 5 0 2E1、谷胱甘肽S 转移酶 1、16 α 碳腈孕烯醇酮诱导的细胞色素P4 5 0酶和S 腺苷 L 甲硫氨酸脱羧酶等。结论 小檗碱的降血糖、抗肿瘤作用以及与其他药物的相互作用可能与一系列代谢相关基因的表达变化有关
Objective To study the effect of berberine on the gene expression profile of rat liver. Methods The rats were intragastrically administered with berberine hydrochloride for 6 days. The liver was extracted and total RNA was extracted. The negative control group and the berberine hydrochloride liver tissue cDNA were labeled with Cy3 and Cy5 fluorescent substances respectively, and the probes were prepared and hybridized with the expression microarray. The fluorescence signal images obtained were scanned by computer and analyzed, and each point was calculated. Cy5 and Cy3 signal intensity ratios. Results Of the 4,096 genes detected, 236 genes were differentially expressed in the berberine hydrochloride group, and 32 differentially expressed genes related to metabolism were identified. Among them, 8 metabolically-related genes were up-regulated, including rat nucleus. Phosphodiesterase, isocitrate dehydrogenase 3, diaphorase and glucose 6 phosphatase. Down-regulated expression of 24 metabolically relevant genes, including uridine diphosphate glucuronyltransferase, cytochrome P4 5 2E1, glutathione S transferase 1, 16 α carbonitrile pregnenolone-induced cytochromes P4 50 enzyme and S adenosine L methionine decarboxylase and so on. Conclusion The hypoglycemic and anti-tumor effects of berberine and its interaction with other drugs may be related to the changes of expression of a series of metabolic-related genes.
