目的探讨姜黄素联合白藜芦醇抑制人头颈部肿瘤细胞系增殖的机制。方法用姜黄素和白藜芦醇联合处理Hep2(人喉癌细胞株)及Fa Du(人下咽癌细胞株),应用MTT(塞唑蓝)比色法检测细胞增殖抑制率,平板克隆增殖实验检测单个细胞集落形成能力,Hoechst33258染色检测细胞凋亡情况,同时应用Real-time PCR法检测Bax和Bcl-2的mRNA表达水平。结果姜黄素和白藜芦醇对Hep2和FaDu细胞增殖有抑制作用,二者联合处理后,对细胞增殖、细胞克隆形成能力及细胞凋亡的抑制均明显增强;同时,姜黄素和白藜芦醇上调细胞中Bax和下调Bcl-2的mRNA水平。结论姜黄素与白藜芦醇联合处理可抑制Hep2及FaDu细胞增殖,作用机制可能与上调Bax、下调Bcl-2基因表达有关。
Objective To investigate the mechanism of curcumin combined with resveratrol in inhibiting the proliferation of human head and neck tumor cell lines. Methods Hep2 (human laryngeal carcinoma cell line) and Fa Du (human hypopharyngeal carcinoma cell line) were treated with curcumin and resveratrol. Cell proliferation inhibition rate and plate clone proliferation The colony forming ability of individual cells was detected by experiments. The apoptosis of cells was detected by Hoechst33258 staining. The mRNA expression of Bax and Bcl-2 was detected by Real-time PCR. Results Curcumin and resveratrol could inhibit the proliferation of Hep2 and FaDu cells. The combination of them inhibited the cell proliferation, cell clonality and apoptosis significantly. At the same time, curcumin and resveratrol Alcohol up-regulates Bax and down-regulates Bcl-2 mRNA levels in cells. Conclusion The combination of curcumin and resveratrol can inhibit the proliferation of Hep2 and FaDu cells. The mechanism may be related to up-regulation of Bax and down-regulation of Bcl-2 gene expression.
