对三组经培养和提纯的不同培养阶段的小鼠腹腔巨噬细胞,用形态学和细胞化学方法,通过细胞内酶活性变化来观察SiO_2对细胞的毒性效应。结果:①SiO_2组细胞吞噬SiO_2粉尘后存活时间较正常组缩短4~5倍。②SiO_2组细胞内酶活性较正常组显著减退,二组间差异显著。③在酶活性动态变化组中实验组细胞内酶活性较正常组为低,二组间差异也极显著。结果表明,细胞内酶活性减退与SiO_2对细胞的毒性效应有关。
To three groups of cultured and purified mouse peritoneal macrophages in different culture stages, morphological and cytochemical methods were used to observe the toxic effects of SiO 2 on the cells through the changes of intracellular enzyme activities. Results: ①The survival time of SiO_2 group after phagocytosis of SiO_2 dust was 4 ~ 5 times shorter than that of normal group. ②SiO2 group intracellular enzyme activity was significantly lower than the normal group, the difference was significant between the two groups. ③ In the group of dynamic changes of enzyme activity, the intracellular enzyme activity of the experimental group was lower than that of the normal group, and the difference between the two groups was also significant. The results showed that the decrease of intracellular enzyme activity was related to the cytotoxic effect of SiO_2.
