ATRA对A549生物学特性及E-CD/catenins表达的影响

目的:探讨诱导分化剂全反维甲酸(ATRA)对肺腺癌A549细胞增殖和E-CD/catenins复合物表达的影响。方法:用细胞生长率分析不同浓度ATRA处理不同时相的A549细胞的增殖特性变化;用免疫组织化学法检测ATRA处理前后A549细胞的E-CD/catenins复合物各成分(E-CD、α-和β-catenin)的表达情况。结果:无ATRA处理组A549细胞增殖力最强,而各处理组细胞增殖能力均有较大降低;免疫组化结果显示无ATRA处理组细胞基本不表达E-CD和α-catenin,而β-catenin主要呈膜表达阳性;处理后的A549细胞E-CD、α-和β-catenin表达水平均有明显增强,并且三者的表达形式均以细胞膜表达为主。结论:ATRA可抑制肺腺癌A549细胞增殖、增强E-CD/catenins复合物表达和膜定位。 AIM: To investigate the effects of all-trans retinoic acid (ATRA) on the proliferation of lung adenocarcinoma A549 cells and the expression of E-CD / catenins complex. Methods: The proliferation of A549 cells treated with different concentrations of ATRA at different time points was analyzed by cell growth rate. The expression of E-CD / catenins in A549 cells before and after treatment with ATRA was detected by immunohistochemistry. And β-catenin) expression. Results: The proliferation of A549 cells without ATRA treatment was strongest and the proliferation of cells in each treatment group was significantly reduced. The results of immunohistochemistry showed that cells without ATRA treatment did not express E-CD and α-catenin, The positive expression of catenin was mainly in the membrane. The expression of E-CD, α- and β-catenin in A549 cells was significantly increased after treatment, and the expressions of all three were mainly expressed in the cell membrane. Conclusion: ATRA can inhibit the proliferation of lung adenocarcinoma A549 cells and enhance the expression of E-CD / catenins and membrane localization.
