利用免疫电镜包埋前与包埋后染色法,分别观察6种布鲁氏菌及耶尔森氏0:9型菌中BsAg的分布情况,发现BsAg主要分布在布鲁氏菌外膜,菌体细胞质内很少,以16M羊种布鲁氏菌株中BsAg抗原最多,其余5种布鲁氏菌株中BsAg较少。用包埋前染色法未发现耶尔森氏0:9型菌外膜表面有BsAg存在,而用包埋后染色法观察到布鲁氏菌体细胞质内及耶尔森氏0:9型菌外膜内侧和菌体细胞质内有微量的BsAg。提示布鲁氏菌细胞质内可能含有少许与耶尔森氏0:9型菌起交叉反应的抗原,表明以BsAg抗原做为布鲁氏菌属特异抗原进行布鲁氏菌与耶尔森氏0:9型菌的血清学鉴别诊断是可行的。
Immunoelectron microscopy before and after embedding staining method were observed in 6 kinds of Brucella and Yersinia 0: 9 bacteria in the distribution of BsAg and found that BsAg mainly distributed in the outer membrane of Brucella, bacteria BsAg antigen is most abundant in brucella strains of 16M sheep and few in the other 5 Brucella strains. The presence of BsAg on the surface of outer membrane of Yersinia 0: 9 strain was not found by the method of pre-embedding staining. However, in the cytoplasm of Brucella bacteria and the Yersinia type 0: 9 strain Inside the outer membrane and bacterial cytoplasm trace BsAg. Suggesting that the cytoplasm of Brucella may contain a small amount of antigens with Yersinia type 0: 9 cross-reaction of antigens, indicating that BsAg antigen as a Brucella specific antigen Brucella and Yersinia 0 : 9 type of serological differential diagnosis is feasible.
