目的评价中药制剂清风胶囊对吗啡依赖大鼠在环境线索诱导下CPP长期表达和觅药行为的影响。方法建立吗啡依赖大鼠条件性位置偏爱(CPP)模型。将大鼠随机分为空白对照组,模型组,阳性药对照组,清风胶囊低、中、高剂量组。清风胶囊低、中、高组给药剂量分别为0.5,1.0,2.0 g/kg,连续给药5天。同时,阳性药对照组给予丁丙诺啡(0.26mg/kg,ip)。分别于实验第16,21,26,31天测定各组大鼠的CPP值。结果模型组大鼠在测试时间内的CPP值均较空白对照组明显增大(P<0.01)。经清风胶囊治疗5天后,大鼠的CPP值与同一时间点的模型组比较明显减小(P<0.01)。阳性药对照组大鼠的CPP值未见明显下降。结论清风胶囊对环境线索诱导的吗啡依赖大鼠CPP长期表达和相关的觅药行为具有抑制作用,并能促进吗啡依赖大鼠的行为恢复正常。
Objective To evaluate the effect of Chinese herb preparation Qingfeng capsule on the long-term CPP expression and drug seeking behavior of morphine-dependent rats induced by environmental cues. Methods The conditioned place preference (CPP) model of morphine dependent rats was established. The rats were randomly divided into blank control group, model group, positive drug control group, Qingfeng capsule low, medium and high dose group. Qingfeng capsule low, middle and high doses of 0.5,1.0,2.0 g / kg, continuous administration of 5 days. Meanwhile, the positive control group was given buprenorphine (0.26 mg / kg, ip). The CPP of rats in each group were measured on the 16th, 21st, 26th and 31st days respectively. Results The CPP value of model rats in test group was significantly higher than that of blank control group (P <0.01). After 5 days of treatment with Qingfeng Capsule, the CPP of rats decreased significantly compared with the model group at the same time point (P <0.01). Positive control group rats CPP value no significant decline. Conclusion Qingfeng Capsule can inhibit the long-term CPP expression and related drug-seeking behavior of morphine-dependent rats induced by environmental cues, and promote the normal behavior of morphine-dependent rats.
