目的:探讨五虎汤对哮喘模型大鼠肺组织α-平滑肌肌动蛋白(α-SMA)表达及气道重塑的影响。方法:呼吸道合胞病毒(RSV)合卵清白蛋白(OVA)致敏激发建立SD大鼠慢性哮喘模型,将48只SD大鼠随机分为正常对照组、模型组、地塞米松组及五虎汤高、中、低剂量组,每组8只。肺组织病理标本行HE染色观察组织病理学改变,Masson染色检测上皮下胶原蛋白含量,免疫组织化学法检测α-SMA表达,并用计算机图像分析系统评价各组气道重塑,并分析胶原蛋白与α-SMA表达的相关性。结果:(1)模型组WAm/Pbm及WAi/Pbm显著高于正常对照组(P<0.01),五虎汤中、高剂量组及地塞米松组WAm/Pbm、WAi/Pbm显著低于模型组(P<0.01);模型组出现炎性细胞浸润增多、上皮细胞脱落、上皮下胶原蛋白沉积及平滑肌细胞层增厚等气道重塑的结构改变,五虎汤中、高剂量及地塞米松可有效减轻上述病理改变;(2)模型组上皮下胶原蛋白及肺组织α-SMA表达显著高于正常对照组(P<0.01),五虎汤中、高剂量组及地塞米松组上皮下胶原蛋白显著表达显著低于模型组(P<0.01),五虎汤各剂量组及地塞米松组肺组织α-SMA表达显著低于模型组(P<0.05或P<0.01);(3)各组大鼠气道上皮下胶原蛋白沉积、α-SMA的积分光密度(IOD值)呈显著正相关。结论:五虎汤中、高剂量能降低哮喘幼年SD大鼠上皮下胶原蛋白及肺组织α-SMA的表达,从而有效抑制哮喘气道重塑。
Objective: To investigate the effects of Wuhu decoction on the expression of α-smooth muscle actin (α-SMA) and airway remodeling in asthmatic rats. Methods: Chronic asthma model was induced in SD rats by sensitization of RSV and OVA. 48 SD rats were randomly divided into normal control group, model group, dexamethasone group and five tigers Tang high, medium and low dose group, each group of 8. The histopathological changes of lung tissue were observed by HE staining. The collagen content of subepithelial tissue was detected by Masson staining. The expression of α-SMA was detected by immunohistochemical method. The airway remodeling was evaluated by computer image analysis system. α-SMA expression of the correlation. Results: (1) WAm / Pbm and WAi / Pbm in model group were significantly higher than those in normal control group (P <0.01). WAm / Pbm and WAi / Pbm in Wuhutang medium and high dose group and dexamethasone group were significantly lower than those in model group (P <0.01). In the model group, structural changes of airway remodeling such as increased inflammatory cell infiltration, epithelial cell detachment, subepithelial collagen deposition and smooth muscle cell layer thickening were observed. (2) The expression of α-SMA in epithelial collagen and lung tissue in model group was significantly higher than that in normal control group (P <0.01), middle and high dose of WuHutang group and dexamethasone group (P <0.01). The expression of α-SMA in the lung of each dose group of WuHuang and dexamethasone group was significantly lower than that of model group (P <0.05 or P <0.01); (3) ) There was a significant positive correlation between the airway epithelium collagen deposition and the integral optical density (IOD value) of α-SMA in each group. Conclusion: Wuhutang medium and high doses can reduce the expression of α-SMA in the sub-dermal collagen and lung tissue of asthmatic SD rats in a dose-dependent manner, which can effectively suppress airway remodeling of asthma.
