发现新的白血病相关基因及探讨白血病的发生机制。方法 :应用分子克隆技术RLGS及RACE技术自急性髓细胞白血病骨髓标本中克隆新的白血病相关基因。结果 :克隆到一个长度为 2 948bp的DNA片段 ,经国际基因库同源检索证明为一未知基因的一部分 ,并定位于第 11号染色体长臂 11区 ,进一步克隆到这一基因的全长cDNA ,其长度为 980bp ,并被国际基因库以新的人类基因LRP16收录入库。结论 :RLGS及RACE技术对于发现白血病相关基因是一种有效的方法 ,LRP16基因的发现对于探讨白血病发生机制具有一定意义。
Discover new leukemia-related genes and explore the mechanisms of leukemia. METHODS: The new leukemia-related genes were cloned from acute myeloid leukemia bone marrow samples using molecular cloning techniques RLGS and RACE. RESULTS: A DNA fragment of 2,948 bp in length was cloned and proved to be a part of an unknown gene by homologous search in the international gene bank. It was located in the 11th region of the long arm of chromosome 11, and the full-length cDNA of this gene was further cloned. Its length is 980bp, and it is included in the database by the International Gene Bank with the new human gene LRP16. Conclusion : RLGS and RACE technology is an effective method for discovering leukemia related genes. The discovery of LRP16 gene has a certain significance for exploring the mechanism of leukemia.
