目的 通过检测肿瘤抑制基因产物PTEN蛋白在肝癌中的表达情况,探讨其作为一种评估肝细胞癌(HCC)生物学特征的分子标记的意义。方法 用免疫组化技术检测3组共43例(孤立性大肝癌、小肝癌及多结节肝癌组)HCC病例与6例正常肝组织中PTEN蛋白的表达。分析HCC中的PTEN蛋白表达与各临床病理特征的关系。结果 PTEN蛋白在正常肝组织及HCC癌旁组织中表达水平高于癌组织,差异具有显著性(P<0.05),但正常肝组织与癌旁组织间表达无显著性差异(P>0.05)。分化程度高的HCC中PTEN表达高于低分化者,无静脉浸润者高于有静脉浸润者,Child A级者高于B级和C级,差异均具有显著性(P<0.05)。PTEN蛋白在大肝癌组表达高于多结节组,但大、小肝癌组间表达无显著性差异(P>0.05)。结论 PTEN蛋白可作为检测肝癌生物学特征的一种分子标记。PTEN检测结果支持孤立性大肝癌较多结节肝癌具有更好的预后。
Explore the significance of evaluating biological characteristics as hepatocellular carcinoma (HCC) inhibit molecular marker gene product PTEN protein expression in HCC tumors by detecting the object. Methods Immunohistochemical technique was used to detect the expression of PTEN protein in 43 HCC cases (isolated large hepatocellular carcinoma, small hepatocellular carcinoma and multiple nodular hepatocellular carcinoma) and 6 normal liver tissues in 3 groups. The relationship between PTEN protein expression and clinicopathological features in HCC was analyzed. Results PTEN protein levels than cancer tissue, the difference was significant (P normal liver and HCC peritumoral tissue in <0.05), but the adjacent normal liver tissue and cancer, there was no significant difference (P expressed interstitial> 0.05). Highly differentiated HCC than PTEN expression in poorly differentiated, without venous invasion were higher than venous invasion were, Child A class by class B and C above, the differences were significant (P <0.05). The expression of PTEN protein in large hepatocellular carcinoma was higher than that in multiple nodules, but there was no significant difference between large and small hepatocellular carcinoma (P> 0.05). Conclusion PTEN protein can be used as a molecular marker for detecting the biological characteristics of hepatocellular carcinoma. PTEN test results support for isolated large hepatocellular carcinoma more nodular liver cancer has a better prognosis.
