应用人肝癌SMMC-7721细胞系和大鼠肝原代培养细胞,利用[3H]TdR掺入的方法观察从人胎肝中提取的肝细胞生长刺激物质(HSS)对肝源性细胞NDA合成的影响。证明HSS对此两种细胞的DNA合成均有显著的促进作用。应用HSS治疗D-氨基半乳糖引起的大鼠肝坏死模型,结果使大鼠的血清谷丙转氨酶有非常显著的下降。本研究进一步证明肝细胞生长刺激因子在人胎肝中存在,并且对肝源性细胞有特异的促生长作用。
Human hepatoma SMMC-7721 cell line and rat primary hepatocyte culture cells were used to observe the effects of hepatocyte growth stimulating substance (HSS) extracted from human fetal liver on the synthesis of NDA from hepatic cells by [3H] TdR incorporation influences. It is proved that HSS can significantly promote the DNA synthesis of these two kinds of cells. HSS treatment of D-galactosamine-induced rat model of liver necrosis, the results of the rat serum alanine aminotransferase have a very significant decline. This study further demonstrates that hepatocyte growth stimulating factor is present in human fetal liver and has specific growth-promoting effects on liver-derived cells.
