目的:探讨T淋巴瘤侵袭转移诱导因子1(Tiam1)及E-cadherin,Vimentin蛋白与上皮间质转化(epithelial and mesenchymal transiton,EMT)在肺腺癌细胞中的表达关系的临床意义。方法:从112例恶性胸水中提取肺腺癌细胞为研究对象,以39例良性胸水中增生上皮细胞为对照,常规液基细胞薄层(TCT)技术、H&E染色、免疫细胞化学染色检测胸水中的肺腺癌细胞。采用免疫细胞化学方法检测细胞中Tram1、E钙粘素蛋白、Vimentin的表达。结果:112例恶性胸水标本中,75例Tiam1在肺腺癌细胞中表达阳性,阳性率为67.0%。E-cadherin及Vimentin蛋白在肺腺癌细胞中的表达率分别为81/112(72.3%)和94/112(83.9%)。结果显示,Tiam1在肺腺癌细胞中的表达的增强伴随E-cadherin表达的下降及Vimentin的表达增强。Tiam1和E-cadherin在增生上皮细胞中阴性表达。结论:Tiam1在肺腺癌细胞中阳性表达,其影响肿瘤发展的机制可能与上皮间质转化发生有关。
Objective: To investigate the clinical significance of the expression of Tiam1, E-cadherin, Vimentin protein and epithelial and mesenchymal transit (EMT) in lung adenocarcinoma cells. Methods: Lung adenocarcinoma cells were extracted from 112 cases of malignant pleural effusion. Thirty-nine cases of benign pleural effusion epithelial cells as control, routine liquid-based cell sheet (TCT) technique, H & E staining and immunocytochemical staining Of lung adenocarcinoma cells. Immunocytochemistry was used to detect the expression of Tram1, E-cadherin and Vimentin in cells. Results: Of the 112 cases of malignant pleural effusion, 75 cases of Tiam1 were positive in lung adenocarcinoma, the positive rate was 67.0%. The expression rates of E-cadherin and Vimentin in lung adenocarcinoma cells were 81/112 (72.3%) and 94/112 (83.9%), respectively. The results showed that the increased expression of Tiam1 in lung adenocarcinoma cells accompanied with the decrease of E-cadherin expression and the increase of Vimentin expression. Tiam1 and E-cadherin are negatively expressed in proliferating epithelial cells. Conclusion: The positive expression of Tiam1 in lung adenocarcinoma cells may contribute to the development of epithelial-mesenchymal transition.
