目的:探讨核心蛋白聚糖(decorin,DCN)对HuH7细胞系体外增殖的影响及机制。方法:加入不同浓度(0、25、50、75、100、125、150、200μg/L)的DCN培养HuH7细胞系不同时间(12、24、48、72h和2周),用二甲基噻唑二苯基四唑溴盐(MTT)法及克隆实验测定细胞增殖速度和活力,流式细胞术测定细胞周期和凋亡情况。结果:DCN浓度增加及作用时间延长,细胞增殖速度减慢、活力减低,G1期细胞增多,凋亡增加。结论:DCN可能为一种负性调控蛋白,可通过调节细胞周期,抑制肝癌细胞的增殖,促进其凋亡。
Objective: To investigate the effect of decorin (DCN) on the proliferation of HuH7 cell line in vitro and its mechanism. Methods: HuH7 cells were cultured with different concentrations of DCN (0, 25, 50, 75, 100, 125, 150 and 200μg / L) for different times (12,24,48,72h and 2 weeks) The cell proliferation rate and viability were measured by diphenyltetrazolium bromide (MTT) assay and cloning experiments. The cell cycle and apoptosis were determined by flow cytometry. Results: The concentration of DCN increased and the action time prolonged, the cell proliferation rate slowed down, the vitality reduced, the G1 phase cell increased and the apoptosis increased. Conclusion: DCN may be a negative regulatory protein that can inhibit the proliferation of hepatocellular carcinoma cells and promote its apoptosis by regulating the cell cycle.
